Thalidomide as a Teratogen

چکیده
Before 1961, there was very little evidence for drug-induced malformations in humans. But in that year, Lenz and McBride independently accumulated evidence that a mild sedative, thalidomide, caused an enormous increase in a previously rare syndrome of congenital anomalies. The most noticeable of these anomalies was phocomelia, a condition in which the long bones of the limbs are absent (amelia) or severely deficient (peromelia), thus causing the resulting appendage to resemble a seal flipper (see Figure 1.16 in Gilbert, 2000). Over 7000 affected infants were born to women who had taken this drug, and a woman need only have taken one tablet to produce children with all four limbs abnormalities induced by the ingestion of thalidomide included heart defects, absence of the external ears, and malformed intestines. The drug was withdrawn from the market in November, 1961.

Thalidomide is the best known human teratogen. Although withdrawn from the market in 1961, thalidomide was remarketed after 1965 in several countries, for the treatment of erythema nodosum leprosum. Thalidomide has a potent immunomodulatory property and has now a number of approved and off-label uses in dermatologic, oncologic, infectious and gastrointestinal conditions. In the U.S., FDA approv...
متن کاملA case of phocomelia associated with diaphragmatic hernia
Objective Phocomelia is a rare congenital anomaly seen in 4 million births. Many factors play a role in the etiology of phocomelia such as environmental-genetic factors, teratogen exposure and amniotic band syndrome. Thalidomide, alcohol, cyclophosphamide and retinoic acid have also been implicated in the pathogenesis. Phocomelia may be isolated or accompanied by syndromes. We present a case of...
متن کاملTeratogen screening using transcriptome profiling of differentiating human embryonic stem cells
Teratogens are substances that may cause defects in normal embryonic development while not necessarily being toxic in adults. Identification of possible teratogenic compounds has been historically beset by the species-specific nature of the teratogen response. To examine teratogenic effects on early human development we performed non-biased expression profiling of differentiating human embryoni...
متن کاملThalidomide is an inhibitor of angiogenesis.
Thalidomide is a potent teratogen causing dysmelia (stunted limb growth) in humans. We have demonstrated that orally administered thalidomide is an inhibitor of angiogenesis induced by basic fibroblast growth factor in a rabbit cornea micropocket assay. Experiments including the analysis of thalidomide analogs revealed that the antiangiogenic activity correlated with the teratogenicity but not ...
متن کاملThalidomide induces limb defects by preventing angiogenic outgrowth during early limb formation.
Thalidomide is a potent teratogen that induces a range of birth defects, most commonly of the developing limbs. The mechanisms underpinning the teratogenic effects of thalidomide are unclear. Here we demonstrate that loss of immature blood vessels is the primary cause of thalidomide-induced teratogenesis and provide an explanation for its action at the cell biological level. Antiangiogenic but ...
متن کاملAcute Pulmonary Toxicity from Thalidomide in a Patient with Multiple Myeloma
There are many factors which determine the drug combination a patient is prescribed when diagnosed with multiple myeloma. Some of these factors include the patient’s age, level of abnormal serum proteins, abnormal metaphase cytogenetics, and whether or not a patient is a candidate for autologous stem cell transplant (1). Thalidomide is a chemotherapeutic drug that acts by inhibiting angiogenesi...
